چکیده

of substrates such as glucose, sucrose, butyric, pentanoic and propanoic acids used as carbon sources affect the accumulation and composition of biopolymers in the cell. The maximum polymer production (0.97 g/L) was obtained when 2.0 g glucose/100 mL was used as a substrate. Accumulation of polymer in the cell was 28% (wt/wt) when pentanoic acid was used as substrate. Characterization of the biopolymers was carried out by using different spectroscopic and thermal techniques such as FTIR, DSC, 1H NMR and GC/MS. Glass transition temperature (Tg) and melting temperature (Tm) of the polymers changed between 4.71-11°C and 147-179.8°C, respectively. Enthalpy of fusion (∆H) varied between 43.82 and 96.70 J/g. Biodegradability of PHB was tested in biological medium and in phosphate buffer solution. Macid Nurbas, and Tulin Kutsal

Cyclic nature of poly(3-hydroxyalkanoate) metabolism in Alcaligenes eutrophus.

The cyclic process of biosynthesis and degradation of poly(3-hydroxyalkanoate) (PHA) was studied in Alcaligenes eutrophus under conditions of nitrogen-limitation of growth. A. eutrophus cells, which had accumulated poly(3-hydroxybutyrate) (PHB) of 55 wt% content within cells from butyric acid, were transferred into a nitrogen-free medium containing pentanoic acid as the sole carbon source and c...

One-Factor-at-a-Time Optimization of Polyhydroxybutyrate Production and Growth of Alcaligenes eutrophus by Altering Culture Parameters and Incubation Time

Polyhydroxyalkanoates (PHAs) are bioplastics derived from renewable resources such as vegetable oils, corn starch, or microbes. The polyhydroxybutyrate (PHB) is a short-chain-length PHA, and the most important bioplastic produced by certain microorganisms in the presence of excess carbon sources. In this study batch cultivation of Alcaligenes eutrophus with the aim of increasing PHB production ...

Regulated expression of the Alcaligenes eutrophus pha biosynthesis genes in Escherichia coli.

A novel poly-beta-hydroxybutyrate (PHB) production system in which the expression and gene dosage of the Alcaligenes eutrophus pha biosynthetic operon were effectively regulated by cultivation temperature was constructed in Escherichia coli. The pha operon was fused to the negatively regulated tac promoter and cloned into a vector in which the copy number is temperature dependent. A two-phase p...

Cloning and expression of the levanase gene in Alcaligenes eutrophus H16 enables the strain to hydrolyze sucrose

Genetic engineering methods were used to enhance the substrate spectrum of Alcaligenes eutrophus H16, a poly-fl-hydroxybutyric acid (PHB) producer. Using parts of the vector pMMB33 and a 2.5 kb DNA fragment of the Bacillus subtilis chromosome a plasmid was constructed bearing the gene for levanase, an enzyme able to hydrolyze various saccharides. After transfer of the levanase gene by triparent...

Cloning of the Alcaligenes eutrophus genes for synthesis of poly-beta-hydroxybutyric acid (PHB) and synthesis of PHB in Escherichia coli.

Eight mutants of Alcaligenes eutrophus defective in the intracellular accumulation of poly-beta-hydroxybutyric acid (PHB) were isolated after transposon Tn5 mutagenesis with the suicide vector pSUP5011. EcoRI fragments which harbor Tn5-mob were isolated from pHC79 cosmid gene banks. One of them, PPT1, was used as a probe to detect the intact 12.5-kilobase-pair EcoRI fragment PP1 in a lambda L47...
